A week after winning his 50th cap in America, Jamie Ritchie has been handed the co-captaincy as Scotland face Chile in Santiago.
The Edinburgh man, who lost the captaincy before the Six Nations, will lead the side along with Sione Tuipulotu.
The pair are among five players retained from the 42-7 win over the US in Washington. Kyle Rowe, Kyle Steyn and Alex Craig also keep their places in the team.
Ritchie shifts to the openside to accommodate Gregor Brown on the blindside flank, while Josh Bayliss takes over at number eight.
Craig is joined in the second row by Ewan Johnson, who makes his first international start, as do props Nathan McBeth and Will Hurd, having made their debuts as replacements earlier in the tour. Dylan Richardson returns at hooker after impressing against Canada.
Behind the scrum, Gus Warr - who scored twice on his debut against Canada - again takes the number nine jersey and partners Ben Healy at half-back.
Tuipulotu is joined in the midfield by his Glasgow Warriors captain Steyn, with Rowe at fullback. Jamie Dobie and Arron Reed - who like Sale colleague Warr marked his Test debut with a pair of tries against Canada - are the wingers.
Edinburgh hooker Patrick Harrison is primed for his Scotland debut from the bench where he is joined by his club colleagues Pierre Schoeman and Javan Sebastian. Glasgow duo Max Williamson and Rory Darge complete the forward reserves.
The matchday 23 is completed by Adam Hastings, Stafford McDowall and Matt Currie.
